You can see in some scenes that the little worm-like shapes at its bottom are human legs. Perhaps the souls that leave the room of Gauf and live a human life are after death absorbed in some way by Lilith. This makes sense considering that it (she) is the key to the TI.

Considering that they are religious, and would disapprove such genetic manipulation in "human beings", probably not.

Maybe the Evas are to represent Belial, Zapan and Ashmodai, three kings of hell. It is said something about their demon-like shape in the appendix of Eva manga volume 1. I remember having read something about demons who had a horn (shogouki), four eyes (nigouki) and a single eye (zerogouki), but I don't remember exactly if they where those I said. But shogouki could be Sammael, Lilith's consort.
